subsequent 20 min. The time to reach maximal incorporation of TdR-3H into the thymidine monophosphate (TMP) pool was slower than into the TDP and TTP pools. In leukemic cells the rate of equilibration of TMP with TdR-3H was related directly to the TdR concentration and inversely to the temperature. With increasing concentrations of TdR, expansion of the TMP pool derived from TdR was greater than expansion of the TDP and TTP pools. In leukemic leukocytes the TDP and TTP pools were approximately equal in size and accounted for 65% of the thymine nucleotides in the presence of 1 ¿IMTdR but only 10% in the presence of 300 ¿IMTdR. In normal leukocytes at these TdR concentrations the TDP and TTP pools accounted for 35% and 5% of the total thymine nucleotides. Over the range of TdR concentrations studied, incorporation of TdR-3H into the TMP pool was 2to 5-fold greater and in corporation into the TDP and TTP pool was 6to 15-fold greater in CML than in normal leukocytes. Incorporation of 3 JUM TdR-3H into DNA was 30-fold greater in CML than in normal

The RNA nucleotide composition in human leukocytes from normal and leukemic cases.

SOME OBSERVATIONS suggest that the RNA-nucleotide-composition of leukocytes from patients with leukemia differs from that of normal cells. Will et al.’ reported that the purine and pyrimidine base contents of total nucleic acids in acute leukemia and in normal bone marrow cells are different. This variation is not caused by any difference in the DNA-nucleotide-composition. In fact, the purine a...

Pyrimidine metabolism in human leukocytes. I. Contribution of exogenous thymidine to DNA-thymine and its effect on thymine nucleotide synthesis in leukemic leukocytes.

The pathways for the formation of DNA-thymine from syn thesis de novo and from exogenous thymidine (TdR) have been studied in intact leukocytes from patients with chronic myelogenous leukemia. The most significant observations were as follows: (a) DNA-thymine derived from exogenous TdR in creases from 13% to 87% over a range of TdR concentrations from 0.03 /¿Mto 300 /XM.(b) TdR causes expansio...

The enzymatic mechanisms for deoxythymidine synthesis in human leukocytes. 3. Inhibition of deoxythymidine phosphorylase by purines.

(1) Synthesis of deoxythymidine by either direct transfer of deoxyribosyl to thymine (pyrimidine deoxyribosyltransferase) or by a coupled deoxynucleoside phosphorylase mechanism is approximately twofold greater with normal leukocyte extracts (55 to 88% granulocytes) than with extracts prepared from leukocytes obtained from patients with chronic myelogenous leukemia. Activities in lymphocytes (n...

Pyrimidine metabolism in man. II. Studies of leukemic cells.

By Lw H. SMITH, JR., FArm A. BAKER AND MARGARET SULLIVAN I HE CENTRAL ROLE of ribonucleic acids and deoxyribonucleic acids in protein synthesis and genetic function, respectively, has led to a continued interest in their metabolism in neoplasia. A number of antineoplastic agents have been developed as structural analogs of purine or pyrimidine precursors of the nucleic acids. These agents have ...
